First when you open TS. Make sure you have "push to Talk" selected for "Voice Send Method" and that you are pushing that key that is selected as your push to talk key when speaking.
Next try going to Settings-->Options and looking at in the "Output Device" is there more then one listed if so try toggling that then going back to local test mode and seeing if it works.
You can try toggleing "WAVE" and "Direct Sound" in the Settings-->Options window and testing in local test mode.
Try going to "Sound and Audio Devices" in your windows control panel (assuming you are using a Windows OS. From there you can go to the "Sound" and "Audio" tabs to see what is selected as your hardware devices.
Test your mic by going to programs-->Accessories-->Entertainment-->Sound Recorder (again assuming you are using windows). Press the Red Circle (record) if you mic is working properly you should be able to canges in the waveform in the window and hear yourself when played back.
